Low-Intensity Focused Ultrasound-Augmented Multifunctional Nanoparticles for Integrating Ultrasound Imaging and Synergistic Therapy of Metastatic Breast Cancer

Abstract The metastasis of breast cancer is believed to have a negative effect on its prognosis. Benefiting from the remarkable deep-penetrating and noninvasive characteristics, sonodynamic therapy (SDT) demonstrates whole series potential leading treatment. To relieve limitation monotherapy, multifunctional nanoplatform has been explored realize synergistic treatment efficiency. Herein, we establish novel nano-system which encapsulates chlorin e6 (Ce6, for SDT), perfluoropentane (PFP, ultrasound imaging), docetaxel (DTX, chemotherapy) in well-designed PLGA core–shell structure. Ce6/PFP/DTX/PLGA nanoparticles (CPDP NPs) featured with excellent biocompatibility stability primarily enable further application. Upon low-intensity focused (LIFU) irradiation, enhanced imaging could be revealed both vitro vivo. More importantly, combined LIFU, exhibit intriguing antitumor capability through Ce6-induced cytotoxic reactive oxygen species as well DTX releasing generate concerted therapeutic Furthermore, this treating strategy actives strong anti-metastasis by lung metastatic nodules significantly reduced. results indicate that SDT-oriented chemotherapy provided promising approach elevating effective suppressing cancer.
